Alcohol is a well-established risk factor for colorectal cancer. This colorectal cancer research project aims to investigate the contributions of alcohol-mediated DNA damage and defective DNA repair pathways to the development and progression of intestinal cancer using both in vivo mouse model systems and cultured colorectal epithelial organoids from mice and humans. The post holder will manage a colony of mice with breeding and genotyping to develop the relevant crosses between mice with defective intestinal alcohol metabolism and inactivated DNA repair mechanisms within intestinal epithelia. Alongside this in vivo work, the post holder will also establish colorectal organoid cultures of epithelial cells both from human colorectal normal and neoplastic samples and from mice with engineered defects in DNA repair genes and alcohol metabolism genes. This will allow experimental analysis of the effects of DNA damage due to alcohol and its metabolites in terms of the cellular consequences including DNA damage, mutations, cell death and neoplastic changes.
Candidates must have a PhD or anticipate completing a PhD in the near future. Laboratory experience and knowledge of basic molecular biology, cell biology and cell culture techniques are required. The position will entail the use of various molecular analytical techniques and approaches, including DNA analysis and sequencing, gene expression analysis, and protein expression studies. An understanding of mammalian genetics and cancer molecular genetics and experience in managing and working with large datasets is also required.
